白芷愈伤组织中总香豆素的提取与测定

摘要/Abstract

摘要： 建立了白芷愈伤组织中总香豆素质量分数的测定方法,同时优选利用超声提取白芷愈伤组织中总香豆素的最佳工艺.采用L9(34)正交实验对乙醇体积分数、超声时间、超声温度以及溶液pH等因素进行研究,确定超声提取的最适条件.以欧前胡素为指标性成分,检测波长为300 nm,利用紫外-可见分光光度法检测白芷愈伤组织中总香豆素的质量分数.结果表明,利用超声提取白芷愈伤组织中总香豆素的最适奈件为:乙醇体积分数为60％,pH为9,超声温度70℃,超声时间30 min.欧前胡素的质量浓度在3.0～30.0μg/mL (R=0.999)内与吸光度呈良好的线性关系,平均加样回收率为98.03％,RSD=2.4％(n=3).该方法操作简单,结果准确,可用于白芷愈伤组织中总香豆素的质量分数测定.利用该方法对白芷愈伤组织和白芷药材进行同时提取测定,结果表明白芷愈伤组织中总香豆素的质量分数是白芷药材中的9.7倍.
