氯化镧和氯化钆对小鼠免疫细胞作用的体外实验

Effects of LaCl_3 and GdCl_3 on Primary Mouse Immune Cells in vitro

摘要: 通过MTT法研究了LaCl_3和GdCl_3对小鼠免疫细胞的作用.结果表明,GdCl_3在0.001~10 μmol/L浓度内均能促进小鼠脾细胞的增殖, 0.001~0.1 μmol/L的LaCl_3促进小鼠脾细胞的增殖,其他浓度没有影响;除0.1 μmol/L浓度外,其他测试浓度下的LaCl_3均促进小鼠T淋巴细胞的增殖;0.001 μmol/L的GdCl_3抑制小鼠T淋巴细胞的增殖,0.01~1 μmol/L时对小鼠T淋巴细胞的增殖没有影响,10 μmol/L时转而促进小鼠T淋巴细胞的增殖.浓度为0.1 μmol/L的LaCl_3和GdCl_3对小鼠B淋巴细胞的增殖有抑制作用,其他测试浓度下均促进小鼠B淋巴细胞的增殖.作用时间为4 h时,0.001 μmol/L的LaCl_3对NK细胞的活性没有影响,其他浓度下的LaCl_3均能提高NK细胞的活性,0.001~10 μmol/L的GdCl_3均能提高NK细胞的活性;作用时间为8 h时,0.001和0.01 μmol/L的LaCl_3显著降低NK细胞的活性,0.1和1 μmol/L的LaCl_3提高NK细胞的活性,当浓度为10 μmol/L时对NK细胞的活性没有影响,1 μmol/L的GdCl_3降低NK细胞的活性,其他测试浓度均能提高NK细胞的活性.这提示LaCl_3和GdCl_3对小鼠免疫细胞的影响模式与其作用浓度、作用时间以及稀土元素的种类都是密切相关的.

Abstract: The effects of LaCl_3 and GdCl_3 on primary mouse immune cells in vitro were studied. The results indicated that GdCl_3(0.001-10 μmol/L) promoted the proliferation of the spleen cells. LaCl_3 had no effect on proliferation of the spleen cells at the concentrations of 1 and 10 μmol/L, but promoted the proliferation of the spleen cells at other concentrations. LaCl_3(0.001, 0.01, 1, and 10 μmol/L) promoted the proliferation of T lymphoid cells stimulated by concanavalin A (ConA). GdCl_3 inhibited the proliferation of T lymphoid cells stimulated by ConA at concentration of 0.001 μmol/L, had no effect on the proliferation of T lymphoid cells at the concentrations of 0.01, 0.1 and 1 μmol/L, but turned to promote the proliferation of T lymphoid cells at the concentration of 10 μmol/L. LaCl_3 and GdCl_3 inhibited the proliferation of B lymphoid cells stimulated by lipopolysaccharides (LPS) at the concentration of 0.1 μmol/L, but they promoted the proliferation of B lymphoid cells at other concentrations. For 4 h, LaCl_3 had no effect on NK cell activity at the concentration of 0.001 μmol/L, but promoted NK cell activity at other concentrations. GdCl_3(0.001, 0.01, 0.1,1, and 10 μmol/L) promoted NK cell activity. For 8 h, LaCl_3 inhibited NK cell activity at the concentrations of 0.001 and 0.01 μmol/L, promoted NK cell activity at the concentrations of 0.1 and 1 μmol/L, but had no effects on NK cell activity at the concentrations of 10 μmol/L. GdCl_3 inhibited NK cell activity at the concentration of 1 μmol/L, but it promoted NK cell activity at other concentrations. The results suggest that the effects of LaCl_3 and GdCl_3 on primary immune cells in vitro depend on time, concentration and their species.
