半仿生法提取夏枯草中熊果酸和齐墩果酸

Study on the extraction of ursolic acid and oleanolic acid of Prunella vulgaris by semi-bionic method

摘要: 分别以水和乙醇为提取溶剂,采用半仿生法提取夏枯草,高效毛细管电泳法(HPCE)测定夏枯草中熊果酸(ursolic acid,UA)和齐墩果酸(oleanolic acid,OA)的含量,并与传统的乙醇提取方法进行比较研究.HPCE法测定条件为:未涂层石英毛细管柱(50 μm i.d.×61 cm,有效长度50 cm);运行缓冲液为25 mmoL/L四硼酸钠(含1.5 mmoL/L的β-环糊精溶液,pH=11);运行电压为14.3 kV;氮气压进样,进样压力19.3 MPa,进样时间3 s;紫外检测波长为210 nm;室温.结果表明:采用HPCE法分析测定夏枯草中UA和OA含量,方法的精密度、准确度良好.传统乙醇提取法中UA和OA质量分数分别为4.116 mg/g和0.700 mg/g,半仿生法乙醇提取液中2者质量分数分别为4.802 mg/g和0.975 mg/g,分别是传统乙醇提取法的1.17倍和1.39倍,而水提取液中2者质量分数较低.半仿生法采用pH 2.0、pH 7.5和pH 8.3的提取液分别提取中药材40、40和60 min,以符合人体胃肠消化转运规律,经实验验证将该法运用于夏枯草中2种三萜酸成分的提取可行.

Abstract: Pulverized prunella vulgaris was extracted with ethanol or water by semi-bionic method. The contents of ursolic acid(UA)and oleanolic acid(OA)were determined by high performance capillary electrophoresis(HPCE)and compared with those by the traditional ethanol extraction method. HPCE with UV detector at 210 nm was applied using a silica capillary column(50 μm×61 cm, effective length of 50 cm). The sample was injected with a pressure injection of 19.3 MPa for 3 s. The separation buffer was composed of 25 mmol/L sodium tetraborate and 1.5 mmol/L β-cyclodextrin buffer(pH 11). HPCE method was performed at room temperature and the runningvoltage was 14.3 kV. The contents of UA and OA in the semi-bionic ethanol extract were 4.802mg/g and 0.975mg/g, which were approximately 1.17-fold(4.116 mg/g)and 1.39-fold(0.700 mg/g)of those in traditional ethanol extraction method, respectively. However, the contents of UA and OA in the aqueous extracts were low. Using semi-bionic extraction method, Chinese herbs were extracted using the solution at pH 2.0 for 40 min, pH 7.5 for 40 min and pH 8.3 for 20 min in turn. Which conforms to the law of the human gastrointestinal digestion and transpcrtions. The results shows that The semi-bionic extraction method could be applied to extract two triterpene acids in Prunella vulgaris.
