香芹酚对米尔伊丽莎白菌的抑菌活性及机制研究

摘要: 米尔伊丽莎白菌(Elizabethkingia miricola)是蛙类“歪头病”的致病病原,对蛙类的健康养殖构成严重威胁.香芹酚具有广谱抗菌的特性,在水产养殖中有作为饲料添加剂的潜在可能.为评估香芹酚应用于防治米尔伊丽莎白菌感染的潜力,测定了其对米尔伊丽莎白菌的抑菌能力,并探究了其作用机制.结果表明:香芹酚对米尔伊丽莎白菌的最小抑菌浓度(minimun inhibitory concentration,MIC)和最小杀菌浓度(minimun bactericidal concentration,MBC)分别为100 μg/mL和200 μg/mL.经香芹酚处理后,细菌菌体可溶性蛋白浓度显著降低,DNA外渗量增加;香芹酚处理导致菌体变形、细胞膜皱缩及破裂,生物膜形成大幅降低,并且生物膜合成相关基因的表达被显著下调.上述结果表明:香芹酚对米尔伊丽莎白菌具有较好的抑菌活性,其通过影响细菌蛋白质的正常合成,破坏细胞膜的完整性以及减少生物膜的形成来抑制细菌的生长.这为香芹酚作为抗菌剂应用于防控蛙类米尔伊丽莎白菌感染提供了参考.

Abstract: Elizabethkingia miricola is the pathogen responsible for frog head tilt disease, posing a serious threat to frog aquaculture. Carvacrol has potential applications as a feed additive in aquaculture due to its broad-spectrum antibacterial properties. To evaluate the efficacy of carvacrol in the prevention and treatment of E. miricola infections, we assessed its antibacterial activity against E. miricola and further explored its mechanisms of action. The results revealed that the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of carvacrol against E. miricola were 100 μg/mL and 200 μg/mL, respectively. After treatment with carvacrol, the concentration of soluble proteins decreased significantly, while the DNA extravasation increased. These findings suggest that carvacrol exhibits strong inhibitory activity against E. miricola by disrupting bacterial protein synthesis, compromising cell membrane integrity, and reducing biofilm formation. This study provides valuable data supporting the use of carvacrol as an antibacterial agent for preventing and controlling frog diseases caused by E. miricola infection.
