Effects of long-term continuous 9.375 GHz microwave radiation on Caenorhabditis elegans

Abstract: The Caenorhabditis elegans was used to explore the effects of 9.375 GHz microwave radiation on the motor ability, reproductive ability and lifespan of Caenorhabditis elegans. The wild-type strair(N2)and sod-3 mutant(gk235)strain of C. elegans were cultured to adulthood and synchronized. The worms were randomly divided into control and radiation groups((1 000±100)worms per group)when- DOI:10.3969/j.issn.1000-1565.2023.06.0089.375 GHz微波长期辐射对秀丽线虫的影响张文丽1,2,李志慧2,董国福2,魏丽亚1,王长振1,2(1.河北大学 生命科学学院,河北 保定 071002;2.军事医学研究院 辐射医学研究所,北京 100850)摘 要:利用模式生物秀丽线虫(Caenorhabditis elegans)探究9.375 GHz微波辐射对其运动能力、生殖能力和寿命的影响.将野生型株系(N2)和sod-3基因突变株系(gk235)秀丽线虫培养至成年期进行同步化处理,同步化后秀丽线虫长至L1期时,随机分为对照组和辐射组,每组(1 000±100)只,对辐射组进行连续不间断辐射(频率9.375 GHz,比吸收率(specific absorption ratio,SAR)4 W/kg,辐射时间24 h/d),直到秀丽线虫寿命结束为止.当辐射到L4期时,定期统计产卵量;辐射第7天和第10天,分别检测各组秀丽线虫的运动能力(头部摆动、身体弯曲和咽部震动次数)和存活时间.结果表明:辐射第7天,与对照组相比,N2株系秀丽线虫的头部摆动、身体弯曲和咽部震动次数均呈下降趋势,但无统计学差异;辐射第10天,与对照组相比,可显著降低N2株系秀丽线虫的头部摆动次数(P<0.05),而身体弯曲次数和咽部震动次数呈下降趋势,但无统计学差异.与对照组相比,长期辐射可显著降低sod-3基因突变株系秀丽线虫的头部摆动次数和身体弯曲次数(P<0.05),而咽部震动次数无统计学差异.与对照组相比,长期辐射对N2 和sod-3基因突变株系的产卵量和寿命均无明显影响.综上,在频率9.375 GHz、SAR 4 W/kg的条件下,长期持续辐射可降低秀丽线虫的运动能力,但对秀丽线虫的生殖和寿命无明显影响.本实验为后续深入研究X波段辐射损伤机体的作用机制提供参考.关键词:9.375 GHz微波;秀丽线虫;运动能力;生殖;寿命中图分类号:Q955;O552.424 文献标志码:A 文章编号:1000-1565(2023)06-0624-07Effects of long-term continuous 9.375 GHz microwave radiation on Caenorhabditis elegansZHANG Wenli1,2, LI Zhihui2, DONG Guofu2, WEI Liya1, WANG Changzhen1,2(1. When C. elegans was radiated to the L4 stage, the number of eggs was counted regularly every day. At the time points on the 7th and 10th day of radiation, the motor ability(the number of head swings, body bends, pharyngeal pump vibrations)and survival time of C. elegans in each group were measured, respectively. The results showed that after the 7th day of radiation, the number of head swings, body bends and pharyngeal pump vibrations of the N2 strain showed a decreasing trend compared to the control group, but there was no statistical difference. After the 10th day of radiation, the number of head swings of the N2 strain was significantly reduced compared to the control(P<0.05), while the number of body bends and pharyngeal vibrations tended to decrease, but they were not statistically different. Long-term radiation significantly reduced the number of head swings and body bends in the sod-3 mutant strain compared to the control group(P<0.05), while the number of pharyngeal vibrations was not statistically different. No significant effect on egg production and lifespan of the N2 and sod-3 mutant strains compared to the control group after radiation was observed. In conclusion, under the condition 9.375 GHz and radiation dose of 4 W/kg, long-term continuous radiation can significantly reduce the motor ability of C. elegans, but it has no significant effect on the reproduction and lifespan of C. elegans. This experiment provides a reference for further study of the mechanism of the X-band radiation damage to the organism.
