    Influence of scour site seismic waveforms on the time-course response of large-diameter variable-section single pile

    To investigate the time-history response of large-diameter variable-section single pile subjected to different seismic waves in scouring sites, vibration table tests were conducted to examine the time-history characteristics of pore pressure ratio in the pile foundation soil layer, pile body acceleration, horizontal displacement at pile top, and bending moment in the pile body. Results indicate that as scour depth increases from 0 cm to 12 cm, the onset time of pore pressure ratio growth exhibits a pre-effect. Acceleration along the pile shaft shows a non-linear, increasing distribution from pile base to pile top, peaking at the pile top. The acceleration response of the pile-soil system is influenced by seismic waveform, soil type, and pile foundation stiffness. During the initial phase of seismic motion, pile-head horizontal displacement increased gradually. It surged sharply once accumulated vibrational energy exceeded the elastic confinement limit of the surrounding soil, subsequently stabilising as energy dissipated. The peak bending moment occurs at the interface between saturated sand and strongly weathered granite. At variable cross-sections, bending moments exhibit significant variations due to abrupt changes in sectional stiffness. Consequently, seismic design for large-diameter variable-section single pile foundations must prioritise enhancing the flexural capacity at soil interface zones and variable cross-sections.

    To address the inconsistency where the incorporation of carbide slag microcapsules(CSM)coated with high-temperature wax into concrete results in increased expansion rate yet decreased strength, this study takes carbide slag concrete as the research object and use a random forest regression model with carbide slag content, curing temperature, curing humidity, and age as input features to simultaneously and accurately predict the compressive strength and restricted expansion rate of concrete. Medium to high carbide slag contents exhibit significant coordination effects in the middle curing stage, with the optimal coordination achieved under 40 ℃ curing conditions. At 60 ℃, the instability of microcapsules intensifies strength loss, while the natural curing environment severely inhibits hydration and expansion reactions, making standard curing the optimal humidity condition. Based on the above findings, precise regulation of concrete strength and micro-expansion performance can be realized by controlling the carbide slag content according to the prediction results under different curing environments.

    The mechanical state of bolted joints in skewed girder sections of steel box girder bridges is complex, posing a key technical challenge in bridge design and construction. Based on the construction project of the prestressed concrete steel box girder bridge over the Beijing-Harbin expressway on the Qinhuangdao section of G102 national highway, simulation analysis and linear monitoring technologies were adopted to obtain the internal stress variation law of high-strength friction bolt groups in skewed girder sections. A mechanical model of high-strength friction bolt groups in skewed girder sections was established using ANSYS finite element software, and the deformation characteristics of skewed girder sections under different connection modes were analyzed, the optimal connection mode was obtained, and the dynamic characteristics of high-strength friction bolt groups were revealed. The dynamic load data show that the most unfavorable positions of the bolt group are the 5th and 15th columns. The numerical simulation results show that under the same load conditions, the maximum shear stress of riveted connections is about 177% of that of high-strength friction bolted connections, and the maximum stress of welded connections is about 440% of that of high-strength friction bolted connections, confirming that bolted connection is the optimal connection mode. The research results provide important scientific basis and engineering reference for the design, construction and monitoring of bolt group structures in steel box girder bridges.

    Dynamics of spiral patterns has been simulated using the Ginzburg-Landau equation. The influences of initial condition, boundary perturbation and boundary shape have been simulated. The simulation result shows that the initial condition and the parameters control the type of pattern; boundary condition incorporates the interplay between the environment and the system. When the symmetry of the boundary conditions is different from the symmetry of the force field, the system state is the result of the two different symmetries interacting. That is the boundary conditions modulate the pattern morphology to some extent. The smaller the area scale, the more obvious the boundary effect.The simulated results match the experiment results. This work is of importance for understanding and controlling spiral patterns.
    Screening of proteins interacting with protein tyrosine kinase LYN

    To refine the signal transduction network mediated by the protein tyrosine kinase LYN and clarify its regulatory mechanism in allergic diseases, this study used a yeast two-hybrid system to screen target proteins that specifically interact with LYN. Self-activation activity and cytotoxicity of the bait protein were verified using haploid and diploid mating assays. Primary positive clones were subjected to sequencing alignment and bioinformatics analysis, and protein-protein interactions were further validated by yeast reversion experiments. A total of 53 potential interacting proteins were obtained in the primary screening. After removing invalid sequences including frameshift mutations, non-coding region translations, and coding region deletions, 10 candidate interacting proteins were retained. Combining bioinformatics analysis and yeast reversion experiments validation, the specific interaction between LYN and polyubiquitin-B precursor(UBB)was finally confirmed.     Effect of baicalin on the inflammatory pathway of macrophages in LM infection

    To investigate changes in the gene expression profile and identify key inflammatory genes and signaling pathways involved in baicalin(BA)intervention against Listeria monocytogenes(LM)infection in murine macrophages, cells were divided into three groups:Control, LM-infected(LM), and BA-treated plus LM-infected(LM+BA). Following treatment, transcriptome sequencing(RNA-seq)was performed. Differentially expressed genes(DEGs)were screened and subjected to protein-protein interaction(PPI)analysis, Gene Ontology(GO)enrichment, and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis. Key findings were validated using reverse transcription-quantitative polymerase chain reaction(RT-qPCR). The results revealed 744 DEGs by comparing the LM group vs. the control group and the LM+BA group vs. the LM group. PPI analysis identified 82 core DEGs. These DEGs were primarily enriched in six inflammatory signaling pathways. RT-qPCR validation confirmed that the mRNA expression trends were consistent with the RNA-seq data. Following treatment, transcriptome sequencing(RNA-seq)was performed. Differentially expressed genes(DEGs)were screened and subjected to protein-protein interaction(PPI)analysis, Gene Ontology(GO)enrichment, and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis. Key findings were validated using reverse transcription-quantitative polymerase chain reaction(RT-qPCR). The results revealed 744 DEGs by comparing the LM group vs. the control group and the LM+BA group vs. the LM group. PPI analysis identified 82 core DEGs. These DEGs were primarily enriched in six inflammatory signaling pathways. RT-qPCR validation confirmed that the mRNA expression trends were consistent with the RNA-seq data.     Elizabethkingia miricola is the pathogen responsible for frog head tilt disease, posing a serious threat to frog aquaculture. Carvacrol has potential applications as a feed additive in aquaculture due to its broad-spectrum antibacterial properties. To evaluate the efficacy of carvacrol in the prevention and treatment of E. miricola infections, we assessed its antibacterial activity against E. miricola and further explored its mechanisms of action. The results revealed that the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of carvacrol against E. miricola were 100 μg/mL and 200 μg/mL, respectively. After treatment with carvacrol, the concentration of soluble proteins decreased significantly, while the DNA extravasation increased.     In this study, the secondary metabolites of fungus Fusarium sp. J3 were isolated and identified to explore novel compounds that possess unique structures and biological activities. The ethyl acetate extract of the rice fermentation of the fungus was purified by separation technologies such as silica gel column chromatography, gel column chromatography and high-performance liquid chromatography(HPLC), and the chemical structures of these compounds were elucidated by NMR and ESI-MS. A total of 17 compounds were isolated, including 1 aromatic compound(14), 1 amide compound(15), 3 terpenoids(16, 22, 28), 3 cyclic peptides(17, 29, 30), 1 ketene compound(18), 5 esters(19, 20, 23, 24, 25), and 3 steroids(21, 26, 27). Among these compounds, 24 was isolated from genus Fusarium for the first time. The anti-inflammatory and anti-tumor activities of these compounds were evaluated. Compounds 17, 24 and 30 showed certain anti-inflammatory effect at a mass concentration of 10 μg/mL, with the inhibition rates against NO reaching about 45%. Finally, comparative experiments are conducted on multiple public datasets to evaluate the proposed method against classic oversampling approaches. The results demonstrate that, compared to methods such as SMOTE-CCKNN, ADASYN-CCKNN, and Borderline-SMOTE-CCKNN, the proposed approach effectively enhances the classification performance for imbalanced data by synergistically combining techniques at the data, algorithm, and model levels.

    To address the issues of strong sampling randomness, low efficiency in random tree expansion, and poor path smoothness in the traditional Informed RRT* algorithm for path planning, an improved Informed RRT* algorithm is proposed. The algorithm adopts a hybrid sampling strategy that combines Gaussian-biased sampling with uniform random sampling to reduce sampling randomness and improve the efficiency of initial path discovery. Bayesian learning is introduced to adaptively adjust the sampling weights and the Gaussian standard deviation. During node expansion, an improved artificial potential field and a greedy step-size expansion strategy are incorporated to enhance the directionality of random tree growth and increase expansion efficiency. DDFE-Net contains two branches based on U-shaped network, namely feature enhancement in low contrast areas(FELCA)branch and wavelet domain feature enhancement(WDFE)branch. The FELCA branch first extracts the complementary features of the image through the dynamic feature extraction(DFE)module, and uses the dynamic multi-head attention Transformer(DMFormer)to capture the global features, to enhance the features of the low contrast region. The WDFE branch first uses the residual module to encode the underlying features of the noisy input image, and then establishes the corresponding relationship between image frequencies in wavelet domain through the dynamic wavelet domain correlator(DWDC)to further increase the spatial continuity of the image. Finally, DDFE-Net fuse the features obtained from the two branches, and then reconstructed a high-quality denoised image. Experimental results show that DDFE-Net is superior to existing methods in noise suppression, edge protection and computational efficiency, especially in dealing with complex noise.
