日本沼虾(Macrobrachiumnipponense) 4个野生群体遗传多样性微卫星分析

Microsatellite analysis of genetic diversity in four wild populations of oriental river prawn(Macrobrachium nipponense)

摘要: 为了评价不同地区日本沼虾(Macrobrachium nipponense)野生群体的遗传多样性和遗传分化水平,利用微卫星标记对白洋淀、衡水湖、微山湖和洪泽湖4个地区日本沼虾野生群体的遗传多样性进行分析.结果显示,22对微卫星位点都显示具足够的多态性;每个野生群体中至少有6个位点显示杂合不足,显著偏离了Hardy-Weinberg平衡;4个日本沼虾野生群体均表现出了较高的遗传多样性,其中微山湖野生群体遗传多样性水平相对较高,其他3个野生群体的遗传多样性水平则相对较低;群体间F-统计量(Fst)及分子方差分析表明,4个日本沼虾野生群体间存在中等程度的遗传差异(0.046 3<Fst<0.101 2);基于Nei’s遗传距离构建的UPGMA系统进化树显示,地理位置相邻的日本沼虾野生群体聚在一起.

Abstract: Genetic diversity and differentiation of different wild population of oriental river prawn(Macrobrachium nipponense)in Baiyangdian Lake,Hengshui Lake,Weishan Lake and Hongze Lake,were investigated using 22 microsatellite DNA loci.The data showed that all the 22 loci were highly polymorphic.An exact P-value test indicated that most loci significantly deviated from Hardy-Weinberg in the Baiyangdian Lake,Hengshui Lake and Hongze Lake populations with the number of 9 loci,respectively;the deviations of Hardy-Weinberg were also detected in Weishan Lake populations with the number of 6 loci.All of the four wild populations showed high genetic diversities,and the genetic diversity in Weishan populations was relatively higher than the others three populations.The Fst and AMOVA analysis suggested that there was a moderate degree of genetic divergence among the four populations(0.046 3<Fst<0.101 2).UPGMA clustering tree based on Nei’s genetic distance demonstrated that the populations of adjacent- geographical position clustered together.
