Direct shoot regeneration in vitro for Atractylodes macrocephala using epicotyl and radicle as explants

Abstract: For establishing the high efficiency regeneration system in vitro of Atractylodes macrocephala Koidz.,the effects of explant types and concentrations of plant growth regulators on the direct shoots regeneration for A.macrocephala were examined.The results indicated that shoots were directly induced from radicle and epicotyl explants without callus induction.The appropriate medium for shoot regeneration was MS + TDZ 1.5 mg/L + NAA 0.2 mg/L.With this regeneration system,91.18% of epicotyl explants produced an average of 13 elongated shoots per initial explant inoculated.The rooting efficiency was up to 66.66% on the 1/2 MS + IBA 0.5 mg/L.The regeneration system for A.macrocephala provided reliable technical support for plant genetic engineering quality improvement,industrial seeding,and virus-free of A.macrocephala.
