Molecular identification of crab spiders from Xiaowutai Mountains,China with DNA barcoding

Abstract: In order to explore the feasibility of DNA barcoding in the species identification of crab spiders,the COI gene of 110 thomisid samples belonged to 25 species in Xiaowutai Mountains,China,was applied; the phylogenetic trees were reconstructed by neighbor-joining(NJ)and Bayesian Inference(BI)methods,and with ABGD(automatic barcode gap discovery)software to divide the samples.The results showed that the samples belonging to the same species clustered in one monophyly and all species were successfully distinguished by the phylogenetic trees,and the groups automatically defined by ABGD perfectly matched the groups defined by morphological characters,and accorded with phylogenetic trees.We believe DNA barcoding can be used as an effective tool in the molecular identification of crab spiders.
