Box-Behnken design for optimized extraction of polysaccharides from maca via ultrasonic-assisted aqueous two-phase method

Abstract: A 17-run Box-Behnken design was used to optimize the extraction conditions of polysaccharides from maca via ultrasonic-assisted aqueous two-phase method.Three factors,namely,extraction temperature,extraction time,and solid-liquid ratio were investigated.The experimental data were fitted to a second-order polynomial equation through multiple regression analysis and examined with the appropriate statistical methods.The adjusted coefficient of determination(R2Adj)for the model was 0.815 3,and the probability value(P<0.000 1)demonstrated a high significance for the regression model.The optimum extraction conditions were as follows:extraction temperature of 60 ℃,extraction time of 52 min,and solid-liquid ratio of 1∶20.Under these conditions,the mean extraction yield of maca polysaccharides was 15.732%,which was in good agreement with the predicted value of 15.831%.Box-Behnken response surface method is used to optimize extraction of polysaccharides from maca via ultrasonic-assisted aqueous two-phase method,the optimized conditions are feasible,the model predition is good.
