Preparation and characterization comparison of FITC bonded fluorescent silica nanospheres with surface connection type and core-shell type

Abstract: Silica fluorescent nanoparticles were prepared with fluorescein isothiocyanate(FITC)as the fluorescent molecule and silica as the biocompatible carrier.To enhance the stability of the microspheres,the bridge molecule(γ-aminopropyl triethoxysilane,APTES)was introduced.Owing to the non-unity of the connection sequences of FITC,SiO2 with APTES,two types(surface connection type and core-shell type)of FITC bonded fluorescent silica nanospheres were prepared and compared in their morphologies and photobleachings.The results show that both types had similar morphology,but the surface connection type was optically bleached seriously.Furthermore,two typical methods of packaging silicon(Stöber and reverse microemulsion)were compared.Core-shell FITC bonded fluorescent silica nanospheres preparaed by reverse microemulsion method showed stronger fluorescence signal,and had smaller particle size and better dispersibility.In addition the value of fluorescent dye leak was less than 8% within 7 h in the dye leakage experiment and the fluorescence intensity decreased only 2.2% within 30 min in light stability experiment,which showed the low dye leakage degree and good light stability properties.-
